Lung Cancer Detection
The Colorado Biomarkers Discovery Laboratory uses microarray technology to detect abnormal patterns of gene expression in lung cancers to identify candidate biomarkers. These are then tested using amplification methods that permit the identification and quantification of aberrent signals in mixtures of tumor and benign cell. At the present time we are focusing on Cancer/Testis genes because of the near complete absence from all somatic human tissue and their high level of expression in cancers.
Lung premalignancy and carcinogenesis
Central cancer is presumed result from a series of stepwise changes in the bronchial epithelium. Better definition of the molecular configuration of these lesion may permit identification of a patient population that could benefit from screening and chemoprevention. We have utilize samples from a 4000 person cohort of smokers to evaluate methylation of tumor supressor genes (Dr Steve Belinsky), SKY FISH and other cytogenetic abnormalities and abnormal gene expression profiles to identify aberrently expressed genes to construct a profile that may predict for the future development of lung cancer. To date a panel of methylation has been developed that has promise for the prediction of lung cancer before over malignancy occurs. SKY FISH may also be predictive. Finally, a profile of premalignant is being developed. These molecular changes are being compared to the current morphological gold standard to develop molecular tests that may supplement and, in some instances, replace morphological biomarkers.
